WHITEFISH, Mont. - A peaceful protest Whitefish, Mont. became the site of a viral confrontation when a man was filmed cursing at protesters and attempting to damage their signs as the demonstrators chanted, "peaceful! peaceful!" Footage of the June 4 incident was published on Instagram. (Warning: video contains foul language.) "This is why we are here," read the caption for the video posted by the Instagram account "charlesnorthwest." The man was identified as 51-year-old Jay Snowden, a resident of the town. Snowden would later be charged with one count of disorderly conduct, according to the Whitefish Police Department. | Study provides molecular explanation to a longstanding mystery in viral oncology - News-Medical.net Posted: 09 Jun 2020 11:47 PM PDT  The oncogenic herpesvirus (HHV8 or KSHV) causes a cancer known as Kaposi's Sarcoma. An international team of scientists led by the University of Helsinki has discovered key factors that control the genome maintenance and replication of a virus responsible for lymphatic vascular cancer. Kaposi Sarcoma (KS) is the most common cancer among AIDS patients and it is often seen in sub Saharan and Mediterranean populations or after graft transplant. Kaposi Sarcoma is suggested to originate from the endothelium in immunocompromised individuals. This disease is caused by a herpesvirus, Kaposi Sarcoma herpesvirus (KSHV). KSHV displays two modes of infection, latency and lytic phase, with both phases contributing to the tumorigenesis. Latency represents the default mode of infection in virtually all susceptible cell types. Quite exceptionally, lymphatic endothelial cells (LEC), the suggested KS tumor cells precursors, display a unique viral infection program with spontaneous lytic gene expression as well as high intra- and extracellular viral load. In the current publication in Cancer Research, a journal of the American Association for Cancer Research, a collaborative team of researchers led by Professor Päivi Ojala decided to address a long-standing question: What is the molecular etiology of the spontaneous lytic phase observed in KSHV-infected primary human lymphatic endothelial cells (KLECs)?
A finding turns around the current dogmaSilvia Gramolelli, an Academy of Finland post-doctoral researcher at the University of Helsinki, investigated the role in Kaposi Sarcoma of PROX1, SOX18 and COUPTF2, the three key transcription factors (TFs) governing the birth of the lymphatic vasculature during embryonic development. The results of the study demonstrate that all three key factors are expressed in a cohort of KS tumors concomitant with unexpectedly prominent marker of the late lytic phase, the protein K8.1. "Key transcription factors SOX18 and PROX1 expressions are crucial to support the spontaneous and productive KSHV lytic infection program in LECs, and their expression in tumors correlates with lytic and latent markers of infection," says Gramolelli. This finding turns around the current dogma that regards the KS tumor cells predominantly latently infected and indicates that lytic gene expression is far more abundant than previously thought. "To our surprise, SOX18 and PROX1 regulate two different and complementary processes in the virus oncogenic replication cycle independently from each other," says Gramolelli. "While PROX1 enhances the viral lytic gene expression, SOX18 is involved in the maintenance of a higher number of viral genome copies. Besides revealing an exciting insight on the KSHV infection program in endothelial cells, this study uncovers how the virus mechanistically hijacks PROX1 and SOX18 to its genome to support its own replication and lytic gene expression," Gramolelli continues. A companion study demonstrating PROX1-Enhanced Productive Lytic Replication of KSHV led by Professor Young Kwon Hong (Keck School of Medicine, University of Southern California) is also published as a back-to-back article in Cancer research. A new KS therapy?Despite decades of research, no standard treatment for KS has been established. Clinical outcomes are particularly unfavorable, especially in resource-limited communities. The research group discovered in collaboration with Associate Professor Mathias Francois (the Centenary Institute, University of Sydney) that both a SOX18 small molecule inhibitor previously developed by Francois and the R-enantiomer of an FDA-approved beta blocker, propranolol, showed a striking reduction in intracellular and extracellular viral loads in vitro. "These findings suggest a potential, novel therapeutic avenue for KS, but further work is required to see if it will be possible to apply them to design new KS therapeutic strategies targeting SOX18," Ojala says. 10 June 2020 Espoo, Finland – Nokia today announced new research highlighting 5G Fixed Wireless Access (FWA) as the most desirable 5G use case amongst consumers globally. The study, which was conducted by Parks Associates, surveyed 3,000 people in the UK, US and South Korea and examined consumer understanding and demand for 5G services across six different use cases including autonomous vehicles, video surveillance and immersive technologies1. The research confirms that there is an opportunity for mobile operators to compete with broadband providers by offering FWA to homes and businesses. The new research highlights that 76 percent of all respondents regard FWA as the most appealing use case overall, with 66 percent claiming they would subscribe to 5G FWA if it cost the same as their current broadband service and delivers the same or better performance. Indeed, more would subscribe if it cost less. Currently, 41 percent of respondents only had the option of a single broadband provider with many resenting this lack of choice. This highlights the opportunity mobile operators have to offer FWA as an alternative to traditional broadband services. The research was conducted prior to the global COVID-19 pandemic, which has driven the world to work and learn from home, however, this has coincided with consumer demand for better quality video calls. Indeed, 90 percent rated high-quality, uninterrupted video streams a "very valuable" aspect of 5G. Additionally, a majority of consumers find 5G video use cases attractive, with 66 percent rating video capture and streaming applications appealing, and 69 percent rating video detection and alerting appealing. More than one-third of consumers found AR experiences for remote commerce appealing even before the COVID-19 crisis. The need and appeal has likely increased with social distancing. More broadly, the outlook for 5G is positive based on the findings. Although current familiarity with 5G remains quite low, with just half of consumers claiming any level of familiarity, the appeal of 5G increases with education. Eighty percent of those very familiar with 5G find it appealing compared to 23 percent of those who are unfamiliar. Further, over 50 percent of smartphone owners said they were likely to switch operator if their current provider doesn't offer them 5G in the next 12 months. Josh Aroner, Vice President of Communication Service Provider Marketing, commented: "Nokia commissioned this research prior to the current global coronavirus pandemic but its insights are still valid and applicable, perhaps moreso in this new environment. While current networks are performing well, consumers are newly appreciating the value of quality networking. "I couldn't believe it," Robert Harrian told The Arizona Republic. "What she said was reprehensible ... I'm very, very sorry." The video, first posted on Facebook by Greg Conn, who was in the Phoenix store, has 20,000 shares, 12,000 reactions and nearly 9,000 comments. It shows Tamara Harrian approaching a customer checking out at the cash register, identified in the post as Karina Rodriguez. The gas station/convenience store was a Shell in north Phoenix, according to the Facebook post. Tamara tells Rodriguez that she isn't welcome in the store and needs to "go back to your country" along with other, similar remarks. As an exchange between the women escalates, Harrian can be seen grabbing Rodriguez's arm. Rodriguez then slaps her across the face. Robert Harrian said he first saw the video when a friend emailed it to him Sunday morning. He says he and his wife aren't racist and that a mental illness can make her act out of character. "I've been brought to tears a lot the past couple days," he said. "I have tried to do my best to share with people what really happened, but the emails don't stop, the posts don't stop." Tamara Harrian could not be reached for comment. Husband's law firm drops to 1 star on GoogleRobert Harrian owns the Harrian law firm in downtown Glendale, which he says received more than 1,000 negative reviews posted online since the video went viral. A former assistant told him that before the video's rise to internet infamy, the law firm had two or three reviews. Before getting some taken down, there were about 1,300 negative ones, he said. Internet users were able to reclassify the law firm on Google's first page as a public restroom and briefly changed the main photo to one of a building with a sign that reads "CLOWN MOTEL" in all caps.  "Yesterday the phone lines were so busy, I couldn't get a call in or out," Harrian said. "So many emails came into me that it crashed our email. When I opened it up this morning, I had 300 emails." He worries his name will be tied to racism and that he'll lose work. He isn't sure his law firm, which employs less than 10 people, will survive. Robert said he and his wife have been members of their church for decades and see the importance of diversity. "I don't have a racist bone in my body," he said. "I'm very, very sorry for what happened ... what my wife said and did and that whole scene. I'm very sorry." 'I was born in America'The video went viral over the weekend when Conn posted it on Facebook. It's since been covered by TMZ and shared by hundreds of Twitter accounts. The fight started when Tamara came into the convenience store "yelling at the clerk to get another clerk to help because the line was getting long," Conn's post says. When another employee came to the cash registers, the white woman told Rodriguez to check out, his post says. As Conn filmed the interaction on his phone, Tamara came up to him and said "you need to leave, you're not a part of this," before going back to Rodriguez and asking her where she was born. "I was born in America, b----, where are your ancestors from?" Rodriguez answers. As Rodriguez and a man with her grab their bag from the checkout counter and head for the door, Tamara tells Rodriguez she's ''going back to Mexico" and grabs her arm, telling Rodriguez to ''keep your hands off me.'' Rodriguez then slaps her in the face. "You deserved it, in my opinion," Conn can be heard saying to the white woman as she leaves. The video does not clearly show whether Rodriguez in some way touched the woman as she claimed, but it's clear the woman was making repeated racist insults to Rodriguez as she was checking out and demanded the clerk not serve her before things became physical. "Racism is alive and well," Conn wrote in his Facebook post. Rodriguez was not immediately available for comment. | Adjust and Unity: Hypercasual mobile games grew 72% in March - VentureBeat Posted: 09 Jun 2020 09:44 PM PDT Hypercasual games, or mobile titles folks can play in a minute or less, grew 72% in the quarter ended March 31 as players turned to mobile games as a salve for the pandemic, according to a report from mobile measurement firm Adjust and game engine maker Unity Technologies. That is just one of the ways that gaming has benefited from the shutdown of everything except online entertainment. People across the globe are spending more time than ever playing games while sheltering in place, with an estimated 2.7 billion users set to spend $77.2 billion on mobile games in 2020, the report said. Hypercasual games are lightweight games that offer a lot of replayability. The report analyzed advertising campaign data across hypercasual games, with the average cost of advertising data from Unity. Hypercasual game sessions grew 72% in March globally. China saw a 300% increase in sessions between December 2019 and March 2020, while South Korea registered a 152% increase. Sessions in Japan and Germany rose by 137% and 69%, respectively. During April and May, hypercasual games remained popular, but the growth rates started tapering off, said Paul Muller, chief technology officer at Adjust, in an interview with GamesBeat. The highest increase happened in China, which grew 3.5 times in the quarter. "We can definitely see that the time spent in games was extraordinary compared to many other kinds of apps," said Muller. "Gaming is definitely the single kind of vertical where you could tell that people are at home, they want to have something to do, and playing something seems like a great way to pass the time. And many of the games have some social component where you can see that there are other people playing too." The report's findings show that hypercasual games saw tremendous growth on mobile in the first quarter of 2020, with app engagement and downloads all jumping to record highs. The report draws on data from the top 1,000 and all apps tracked on the Adjust platform, and hyper casual ad campaigns across Unity's network between Q4 and Q1 2020. The strength of hypercasual and mobile games during the pandemic has proven that the smartphone is often the preferred device for a lot of people, even when they are home and they have access to laptops and desktop computers or game consoles, Muller said. "We can see that people actually use the phone because it has over the years become the primary computing interface," he said. "It's the logical one to turn to when you want to pass some time doing something. In 2010, you would use your computer. But in 2020, you use the phone to play something. The pandemic gave us an accelerated view of that." On the lower end of growth, U.S. sessions increased by 35%, and rose 34% in the U.K. Because hypercasual users in the U.S. are already heavy gamers — contributing to 15% of the global sessions based on an average from December 2019 to April 2020 — the increase was not as large. Muller said that cultural differences and different user behaviors may have made a difference in the different growth rates in the U.S. compared to China. While overall costs per install (CPIs, or the cost of advertising a game to get one person to install it) decreased 35% by the end of March, the cost of acquiring hypercasual users in the U.S. is more than double that of Europe and the Middle East. Nevertheless, marketers can expect the highest conversion rates (17%) from the U.S., the gold standard of hypercasual mobile gamers. At the same time, costs in Asia Pacific declined sharply, at around 20 cents. "Clearly one thing that happened in the pandemic was the cost per install has fallen," Muller said. "We're seeing some now we're seeing a bit of a bounce back in the last two weeks, as costs are recovering to a more pre-pandemic level, but it's a little bit too early to say where it will actually level out." Globally, CPIs are much lower, averaging 17 cents in Q1 2020. Because of users' openness to ads, hypercasual games have been able to build a unique business model for mobile gaming, relying on ad monetization for 95% of their revenue. Hypercasual games are taking a significant chunk of the overall advertising for games, Muller said. Muller said that hypercasual games blend the best ad experience and the most engaging mechanics to appeal to the broadest set of players. As such, they have redefined how to do user acquisition and ad monetization. Because of the short-lived appeal of each of these games, users are continuously driven to the next app within a company's portfolio — creating a snowball effect, as users roll and grow the company's user base. "One of the things that we see with hypercasual games is that they have a different approach to classic gaming publishers," Muller said. "It's not about publishing one game and having a successful user base and nurturing that. Rather, it's a short-life game. The people stick around significantly less than for a casual game. The goal is not to get someone to play one game for a long time. The goal is to get you to install and like five or six of my games. And so, overall, you will spend roughly the same time across the games and they will actually monetize a little bit better because of the extremely aggressive monetization." Agatha Hood, head of global advertising sales at Unity, said in a statement that hypercasual game developers have become masters of creating effective ads. these ads have high click-through and install rates at 22% and 14%, respectively. As people stay inside, engagement is up but so is competition for the attention of users. As the cost of advertising is going back up, Game companies may soon have to get used to the idea of spending more on their mobile game marketing campaigns compared to the past few months, Muller said. "It's definitely time to make sure that you have your strategy in place," he said. That was the message posted to Facebook on Tuesday evening on behalf of Days of Our Lives actress Judi Evans. In the post, a photo showed a hospitalized but smiling Evans holding a note with the message on it. The soap star's note came in response to an outpouring of well-wishes from fans. According to her representative, Evans was admitted to the hospital after a horse-riding accident and, while there, became infected with COVID-19. From her representative, Howie Tiger Simon: I spoke to Judi on Sunday and she is STILL in the hospital — 23 days now and counting. She contracted COVID-19 while there and she had what is known as the COVID blood clots in her legs and she nearly had both legs amputated on two different occasions. She had mild symptoms — fever, aches, a cough and the blood clots. On top of everything, when she went into surgery on one of her legs, they forgot to numb the leg and cut into her leg while she was fully (conscious) with no numbing of the area!
